Vijay Kumar NallaniParticipantDear all,
We are planning to grow small volumes (1-3ml) of cell cultures in the deep well plates that are also suitable for small-scale transfections. Can anyone recommend any particular deep well plates with lid or film from any company? Your suggestions are highly appreciated!
Thank you!
Kind regards,
Vijay. -

Bjørn VoldborgParticipantAccording to my Cell Technician, we use deepwell plates from EnzyScreen,
https://www.enzyscreen.com/microplates_96_well_mtps.htm
but according to her, the cells do not grow well in the full deepwell plates. Henning Gram (the author of the paper) was pretty fond of the half-deep well plates, so they may be worth trying out.

Andrea ValaParticipantHi Vijay,
We do 1 ml cultures in 96 well plates and 4 ml in 24 well plates. I don’t know exactly the brand (will have to check the next time I am in the lab – mainly working from home these days).
But one important think is to test growth and viability of your cells in the plates you have. We test cells alone and compared growth/expression of standard protein with growth in 125 ml flask, but also compare transfected cells in the conditions we think are best and compared to expression in a 125 ml flask).
We saw a big difference between round and squared wells, volume per well (eg more than 5 mL in 24 well plates we tried was a no no), and which shaker you use (and the diameter of rotation). Also, this is cell line dependent, so we do optimize these parameters every time.
Hope this is a useful.
Best, Andrea

Vijay Kumar NallaniParticipantAre you talking about this 24 well plate, Andrea?
We would like to grow 1-3 ml suspension cultures. That’s right, it depends on the shaker etc. Is the shaking speed 130 rpm okay in this kind of plates without settling down the cells to bottom of the tube? Probably we need to optimize this.
Which is the best in your case – round or squared well plates? It would be great if you could just let me know the brand whenever it is possible.
Thank you,
Best,
Vijay. -

Andrea ValaParticipantHi Vijay,
we use a different 24 well plate (but don’t know the specifics at the moment since I normally don’t run the experiments myself). Also, will find out the square vs round etc
I should be back at work next week (since little kids are allowed to go back to school) and will have time to find that info for you.
Can you send me your email?
Best, Andrea
