Jan BlahaParticipantDear P4EU members,
I was wandering if anyone adapted and optimized the TetON inducible piggyBac system (Rini et al. 2013) for generation of stable cell lines in HighFive cells? Do you have any experience with sensitivity of HighFive to doxycycline?
Thanks

David HackerParticipantHello Jan,
As HighFive cells are from a lepidopteran species the genome should already contain functional piggybac transposons, making these cells incompatible with the use of an exogenous piggybac transposon. We have used the piggybac system for the generation of stable S2 cells. However, we have only used a constitutive promoter. I can provide further information, if needed.
Best regards,
